The biggest changes are lens correction and the ability to upload to photo sites like flickr directly from Lightroom. There are also improvements to the RAW converter and noise reduction as well as umpteen small improvements to the user interface. With version 3, Lightroom will now manage your video in the same interface but obviously without the editing capabilities.
You can download a 30 free trial of Lightroom 3 to gauge if the upgrade is worth it for you. Remember to back up your Lightroom 2 catalogues before you upgrade them.
For me, the lens correction has meant that I now depend much less on DxO than before, although DxO still offers the superior morphological conversions.
Another useful bit of functionality is that you can set the max file size output when burning jpegs, this comes in handy with flickr, for example, that has a 20 Mb size limit per photo.
